Abstract
Introduction: Doctors who work in the operating room have a higher risk of deoxyribonucleic acid damage related to low vitamin D levels due to lack of sunlight exposure and high exposure to the inhalation anesthetic sevoflurane. This study aims to determine the association between vitamin D (25[OH]D) levels and deoxyribonucleic acid damage in terms of γ-H2AX foci, lymphocyte micronucleus frequency, and 8-OH-dg levels. Method: A cross-sectional study was conducted on 47 subjects in operating room of RS. Dr. Moh. Hoesin (RSMH) Palembang. Doctors aged 25-50 years, working in the operating room at least 20 hours/week, willing to take part in the study and sign the consent were included in the study; but those who were pregnant, with smoking habit (³11 cigarettes/day), alcohol drinker (³3 times/week), with a malignant disease, and chemotherapy or radiotherapy history were excluded. Subjects who withdrew were dropped out. Cutt off used in vitamin D deficient was 25 ng/ml. Data analysis includes descriptive and inferential analysis. Inferential analysis includes numerical comparative using independent t-test/Mann-Whitney and correlative Pearson/Spearman analysis. Result: Twelve subjects were with vitamin D deficiency and 35 subjects were not. The median of DNA damage parameters examined were higher in the vitamin D deficiency group but not statistically significant: γ-H2AX foci 2 (0-8) vs 1 (0-12); p = 0.803, lymphocyte micronucleus frequency 5.083 ± 0.633 vs 4.823 ± 0.463; p = 0.766, and 8-OHDG levels 45.93 (9.11-102.01) vs 36.68 (9.09-476.44); p = 0.643. There was a not significant negative correlation between vitamin D levels and γ-H2AX foci (r = -0.121; p = 0.416), lymphocyte micronucleus frequency (r = -0.171; p = 0.254), and 8-OH-DG levels (r = -0.191; p = 0.197). Conclusion: There was no association between vitamin D levels and γ-H2AX foci, lymphocyte micronucleus frequency, and 8-OH-dg levels.
