Abstract

Pomegranate (Punica granatum L.) peel has been proven to contain secondary metabolites, one of which is the phenolic group in the form of ellagic acid which has potential ability as a treatment for hyperpigmentation. The deformability and elasticity of transethosomes can penetrate the stratum corneum and increase the transfer of active substances in skin permeability. This study aims to determine the optimum formula of transethosomes from variations of 96% ethanol concentrations of 20% and 40% with tween 80 concentrations of 0.5% and 0.75%. Pomegranate peel extract was extracted using the USG Assisted Extraction (UEA) method. Transethosomal nanovesicles were encapsulated using the thin layer hydration method. Optimization was carried out using the 22 factorial design method using the Design Expert 12® program against the response of encapsulation efficiency values, zeta potential, particle size and polydispersity index. The optimum formula was characterized in the form of encapsulation efficiency, zeta potential, particle size, polydispersity index, %EE decrease and pH decrease, then the optimum formula was carried out using one sample t-Test. The total phenolic content of pomegranate peel extract was 155.495 mg GAE/g with a percentage of 15.5495%. Based on the Design Expert 12® analysis, the optimum transethosome formula was obtained at an ethanol concentration of 40% and tween 0.511% with an encapsulation efficiency value of 93.91%, particle size of 157.36 nm, zeta potential -19.73, Polydispersity Index 0.257, %EE decrease 2.403 and pH change 0.33.
