Abstract
Jackfruit leaves (A. heterophyllus) contain flavonoid compounds that are active as anti-inflammatory. Anti-inflammatory is related to antihyperuricemia. This study aims to evaluate the antihyperuricemia activity of the ethyl acetate fraction of jackfruit leaves in male white rats of the Wistar strain. The ethyl acetate fraction was obtained from extraction by a cascading maceration method starting from n-hexane solvent followed by ethyl acetate solvent. For the ethyl acetate fraction, the extract was standardized and antihyperuricemia activity was tested using 35 mice consisting of 7 groups, namely the normal group, the negative control group (Na-CMC), the positive control group (allopurinol), and the treatment group with doses of 100 mg/kgBB (P1), 200 mg/kgBB (P2), 400 mg/kgBB (P3), and 800 mg/kgBB (P4). To get hyperuricemia, rats were given chicken liver juice and purine rich melinjo seeds. Uric acid level measurements were carried out on days 0, 15, 20, 25, and 30. The standardization results showed that the ethyl acetate fraction of jackfruit leaves met the parameters of the extract requirements. The results of the antihyperuricemia test f ethyl acetate action showed antihyperuricemia activity and the highest activity was shown by the treatment of a dose of 800 mg/kg BB with a % decrease in acid content of 79.37%. For positive control, allopurinol provides antihyperuricemia activity with a % decrease in uric acid levels of 90.72%. Statistical analysis showed that there was no significant difference (P > 0.05) between the 100 and 200 mg/kg BB dose treatment but there was a significant difference (P < 0.05) with the 400 and 800 mg/kg BB dose treatment. The analysis also showed that there was a significant difference between the treatment dose of 800 mg/kg BB and the allopurinol-positive control group. The ethyl acetate fraction showed an effective dose value (ED50) of 348,598 mg/kgBB. This data shows that the ethyl acetate fraction of jackfruit leaves has the potential to be developed as a source of raw materials for uric acid
