Abstract

Introduction: Endometriosis is characterized by abnormal tissue growing outside the uterine cavity. The EGFR gene is a proto-oncogene involved in cell proliferation, division, and development, which, under normal conditions, is expressed at relatively low levels in the endometrium. When EGFR is overexpressed, it can lead to signaling disturbances, resulting in excessive cell proliferation. This study investigates the relationship between the status and level of DNA methylation of the EGFR gene promoter and endometriosis using menstrual blood. Methods: This study is an observational analytic research with a case-control approach. MS-PCR was performed on 20 subjects in both the case and control groups. Pyrosequencing was then conducted on 14 samples from each group. Results: The Chi-square test results on DNA methylation status and endometriosis showed no significant relationship (p>0.05). Meanwhile, analysis of DNA methylation levels and endometriosis revealed a significant relationship with a p-value of 0.033 and an odds ratio of 13. Conclusion: There is no correlation between the DNA methylation status of the EGFR gene promoter and the occurrence of endometriosis. However, there is a correlation between the DNA methylation levels of the EGFR gene promoter and the occurrence of endometriosis. Keywords: DNA methylation, EGFR, endometriosis, epigenetic
