Abstract

Sukun (Artocarpus communis Forst) is a plant from the Artocarpus genus in the Moraceae family, commonly found in tropical regions such as Malaysia and Indonesia. Traditionally, this plant has been widely used for medicinal purposes, including as a remedy for fever, diabetes, and toothache. The plant is known to be rich in flavonoid compounds, particularly in the stem and root parts, but information on the flower has not been extensively reported. Therefore, the objective of this study was to isolate phenolic compounds from the methanol extract of breadfruit flowers and test their antibacterial and cytotoxic activities. The isolation process was carried out through maceration extraction using methanol as the solvent. The compounds were then separated using various chromatographic techniques, including fast column chromatography (FCC), Sephadex column chromatography, and pipette column chromatography. The purity of the isolated compounds was tested using thin-layer chromatography (TLC) with several variations of eluents and characterized using UV-Vis and IR spectrophotometry. Antibacterial testing was performed against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) using the disk diffusion method, while toxicity testing was conducted using the BSLT (Brine Shrimp Lethality Test). The process of isolation of compound is resulted light yellow oily solids weighing 6 mg. UV spectroscopy showed peaks at absorbances of 210 nm, 226 nm, 273 nm, and 296 nm. IR spectra showed stretching vibration O-H (3317 cm-1), C-H stretching (2943 and 2831 cm-1), isolated double bond (1644 cm-1) and (1449 cm-1) is the bending vibration of the methyl group, and C-H (620 cm-1). Antibacterial activity of the isolated compounds was tested at concentrations of 2000 ppm, 1000 ppm, 500 ppm, 250 ppm, and a positive control of chloramphenicol (2500 ppm). The antibacterial test results indicated that the isolated compounds fall into a moderate activity category. For S. aureus, the average inhibition zone diameters were 7.68 ± 0.54 mm, 7.21 ± 0.29 mm, 7.01 ± 0.68 mm, and 6.62 ± 0.70 mm, with inhibition percentages of 30.54%, 28.67%, 27.88%, and 26.33%, respectively, compared to the positive control. For E. coli, the average inhibition zone diameters were 9.28 ± 1.49 mm, 6.66 ± 0.32 mm, 4.59 ± 3.41 mm and 6.89 ± 0.88 mm, with inhibition percentages of 39.93%, 28.14%, 19.66%, and 29.52%. The toxicity test revealed an LC50 value of 881.657 mg/L or has a low toxicity level.
