Abstract

The Kardia plant (Bellucia Pentamera Naudin), a member of the Melastomataceae family, contains compounds such as steroids, triterpenoids, and flavonoids, which exhibit potential analgesic and anti-inflammatory properties. This study aims to characterize the extract and evaluate its analgesic and anti-inflammatory effects based on the reduction in writhing behavior in mice and edema volume in rats treated with extracts and fractions of Kardia leaves. The analgesic and anti-inflammatory tests were conducted using the writhing test and rat hind paw edema method on 24 male Wistar rats, divided into six groups: a positive control group treated with sodium diclofenac, a negative control group treated with 1% Na CMC, and test groups (ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction) at a dose of 400 mg/kg body weight. The highest analgesic and anti-inflammatory activity was observed in the n-hexane fraction, which contained triterpenoid and steroid compounds. The highest analgesic percentage was recorded in the n-hexane fraction (73.74%), followed by the ethyl acetate fraction (71.74%), ethanol extract (67.99%), and ethanol-water fraction (64.16%). The n-hexane fraction group showed no significant difference compared to the positive control group (72.44%). The highest anti-inflammatory inhibition percentage was also observed in the n-hexane fraction group (51.20%), followed by the ethyl acetate fraction (50.44%), ethanol extract (50.00%), and ethanol-water fraction (41.00%). The n-hexane fraction group showed no significant difference compared to the positive control group (61.00%).
