Abstract

Cefixime is a third-generation cephalosporin antibiotic known for its low solubility and permeability. Various technologies have been developed to address this issue, one of which is solid dispersion technology. In this study, chitosan and sodium alginate were used as carrier matrices to produce cefixime solid dispersions. The aim of the study was to evaluate the effect of chitosan and sodium alginate variations as polymers on the dissolution rate of cefixime solid dispersions using the co-grinding technique. The co-grinding process was carried out using a planetary ball mill, which simultaneously mixed the active ingredient and the carrier matrix. This technique was chosen because it effectively transforms the crystalline phase into an amorphous form. The formulation of cefixime solid dispersions was prepared with three variations of sodium alginate: 200 mg, 250 mg, and 300 mg. Sample characterization was conducted using XRD, FTIR, and SEM tests, solubility testing with UV-Vis spectrophotometry, dissolution rate testing, and flow properties evaluation using a flow tester and tap density tester. The results showed that Formula 2 of the cefixime solid dispersion was the best formulation, as it reduced the degree of crystallinity to 21.71%, and FTIR analysis did not indicate the appearance of new peaks. The evaluation of cefixime solid dispersions showed changes in particle morphology. Additionally, there was an increase in transmittance percentage in SIF to 98.587 ± 0.019, an increase in the dissolution rate of cefixime to 83.61%, an increase in flow rate to 6.3 ± 0.14 g/s, an angle of repose of 26.4 ± 0.4°, a compressibility index of 16.3 ± 0.29%, and a Hausner ratio of 1.19 ± 0.35. Based on these results, it was concluded that the co-grinding method using chitosan and 250 mg sodium alginate variation was effective in improving the solubility and dissolution rate of cefixime.
