Abstract

A. mangium is one of the plants grown in Industrial Plantation Forests in Indonesia. This plant is one of the plants that has many uses. However, in 2011 wilt disease caused by Ceratocytis fimbriata was reported in Riau. In 2019 this disease was reported in South Sumatra. Therefore, research on morphological and genetic characteristics as well as Koch's postulate test and host range test of the cause of wilt disease in A. mangium plants and tests are needed. This study aims to: 1). To determine the morphological characteristics of pathogen isolates that attack A. mangium, 2). To determine the genetic diversity of pathogen isolates that attack A. mangium 3). Knowing the host range of pathogens that attack A. mangium. Then Koch's Postulate Test and Host Range Test were conducted. Surveys and sampling of diseased plants were carried out in several locations such as Ogan Ilir Regency, Ogan Komering Ilir Regency and Palembang City. Samples were taken from plants that showed symptoms of the rate. The part of the plant taken is the sapwood part that has black spots. Samples taken will be isolated on MEA media for morphological identification both microscopically and macroscopically. As well as molecularly identified using two beta genes βT1a and βT1b. and also using internal transcribed spacer (ITS) namely ITS1 and ITS4 with ITS1 and ITS4. Koch's Postulate Test and Host Range Test were then conducted. The survey results at the three locations showed different incidence levels from 2019 to 2023. Disease attack in OKI in one location with disease attack incidence from 2019-2023 is 25% to 63.15% increased to 38.15%. The attack in OI occurred at Sriwijaya University reaching 100% with an increase of 16.7%, while the attack in Palembang City at one point in Jakabaring with an incidence of 29.03%-87.1% which increased to 58.07%. Macroscopic identification shows that each isolate has the same color, Olive gray, with moderate mycelial growth and Irregular form and Undulate margin. Microscopically each isolate comes from the genus Ceratocytis and has peretecium, chlamydospores, ostiolar hyphae, cap- shaped ascospores. ITS results identified two haplotypes, ITS5 and ITS7, of Ceratocystis fimbriata sensu stricto, and using β-tubulin placed it in the Latin American Clade (LAC) of C. fimbriata. Based on Koch's Postulate test and host range test, the lesion length found on A. mangium seedlings was 7.41 - 11.2 cm. Artocarpus heterophyllus showed lesion length between 6.94 - 7.96 cm. Annona muricata seedlings used produced lesion lengths of 10.76 - 12.56 cm. While in Lansium domesticum the lesion length ranged from 8.74 -14.76 cm and in Mimusops elengi seedlings the lesion length ranged from 7.69 - 11.14 cm. The percentage of plants that died after Koch's postulate test and host range test reached 100%. Keywords: Ceratocytis fimbriata, Ceratocytis wilt disease, Industrial plantation forest, Acacia.
