Abstract

This study aimed to evaluate the antibacterial activity of ethanol extract and ethyl acetate fraction of kenikir leaves (Cosmos caudatus Kunth.), which contain secondary metabolites such as flavonoids, tannins, and saponins. These compounds are known for their potential to inhibit the growth of Propionibacterium acnes and Pseudomonas aeruginosa. The antibacterial activity was assessed using the disc diffusion method to measure the inhibition zone. MIC, MIC50, and MBC values were also determined for both extracts and fractions against the pathogenic bacteria. Characterization of the ethanol extract showed water content of 11.2 ± 1.6 g/mL, drying shrinkage of 6.33 ± 1.53%, water-soluble extract content of 54.3 ± 2.5%, ethanol-soluble extract content of 63 ± 2.0%, and total ash content of 4.93 ± 0.35%. The antibacterial tests were divided into two groups: ethanol extract and ethyl acetate fraction, each tested at concentrations of 25%, 50%, 75%, and 100%, along with positive and negative controls. The best results were observed in the ethyl acetate fraction at 100% concentration, showing inhibition zones of 18.3 ± 1.5 mm against P. acnes and 19.3 ± 1.5 mm against P. aeruginosa. MIC was 25 mg/mL, MIC50 values were 4.84 mg/mL and 9.08 mg/mL, and MBC was 50 mg/mL.
