Abstract

A degenerative disease is a health condition that causes damage or a gradual decline in the function of the body's organs and tissues. One of the main factors that trigger degenerative diseases is the presence of free radicals, which are reactive molecules that can damage body cells. To combat the adverse effects of free radicals, the body needs antioxidants, which are compounds that can neutralize free radicals and protect cells from damage. One source of natural antioxidants comes from the puspa plant (Schima wallichii (DC) Korth). The purpose of this study was to determine the IC50 value of each pure compound and the class of compounds contained in puspa leaves (Schima wallichii (DC) Korth). Determination of antioxidant activity of puspa leaves was carried out qualitatively (KLT) and quantitatively (UV-Vis Spectrophotometer) using the DPPH method. The results showed that the IC50 values of n-hexane, ethyl acetate and methanol-water fractions of puspa leaves were 39.27 ppm; 68.63 ppm and 223.67 ppm, respectively. The methanol-water fraction has weak antioxidant activity, so it is not continued at the next stage. The total pure compounds obtained from puspa leaves after the KCV process and column chromatography are 5 isolates given the code N1, N2, N3, E1 and E2 with IC50 values of 19.47 ppm; 89.64 ppm; 127.17 ppm; 61 ppm and 104.54 ppm, respectively. The content of secondary metabolite compounds owned by puspa leaves, terpenoids contained in isolates N1, N2 and E1, tannin contained in isolates N1, N3 and E2.
