Abstract

Free radicals generated by the body due to environmental factors such as pollution, UV rays, and stress can cause oxidative stress that contributes to degenerative diseases such as cancer and cardiovascular disease. Antioxidants can help neutralize free radicals, and natural sources from plants are considered safer than synthetic antioxidants. One source of natural antioxidants comes from the leaves of Hypserpa nitida. This study aims to identify the class of compounds contained in Hypserpa nitida leaves and determine the activity of antioxidant compounds contained in Hypserpa nitida leaves. This research was conducted from August to December 2024 at the Genetics and Biotechnology Laboratory, Department of Biology, FMIPA, Sriwijaya University. The research methods used are making simplisia, extraction, fractionation, testing the antioxidant activity of fractions using thin layer chromatography and UV-Vis spectrophotometer methods, purification of compounds, testing the activity of pure compounds using KLT and UV-Vis spectrophotometer, and determining the IC50 value with the DPPH method. The results showed that the IC50 values of n-hexane, ethyl acetate and methanol-water fractions of Hypserpa nitida leaves were 36.27 ppm; 71.58 ppm and 265.32 ppm, respectively. The methanol-water fraction has very weak antioxidant activity, so it is not continued at the next stage. The total pure compounds obtained from Hypserpa nitida. after the KCV process and column chromatography are 5 isolates given the codes N1, N2, N4, E1 and E2 with IC50 values of 18.30 ppm; 75.75 ppm; 207.84 ppm; 60.07 ppm and 70.27 ppm respectively. The content of secondary metabolite compounds owned by Hypserpa nitida, steroids found in isolates N1 and N4, flavonoids found in isolates N2 and E2 and terpenoids found in isolate E1.
