Abstract

Myocardial infarction and stroke are the most serious cases of thrombosis and contribute to the global mortality rate. Clinically, thrombosis management therapy is generally given antithrombosis in the form of anticoagulants, antiplatelets, and thrombolytics. Black soybean tempeh is a food fermented with Rhizopus oligosporus fungus. The fungus is rich in proteolytic enzymes, which have strong potential as thrombolytics. The enzyme extract of black soybean tempeh has been studied to have thrombolytic compound activity that will break down thrombus. The purpose of this study is to obtain the optimum liposome formula that can deliver enzyme crude extract and black soybean tempeh purified enzyme to the target thrombus efficiently and safely with the results of liposome characterization including size, PDI, zeta potential, TEM, cycling test, photothermal efficiency, and thrombolytic activity. The experimental design used in this study was a 22 factorial design, namely, 2 factors affecting 2 levels. The factors used were soy lecithin and tween 80. The levels used were minimum and maximum levels. The concentration variations used were soy lecithin (0.8 and 0.1%) and tween 80 (0.1 and 0.01%). The optimum formula obtained was soya lecithin concentration 0.6% and tween 80 concentration 0.01%. The optimum formula produced spheric liposomes with a particle size of 607.8 nm; PDI value of 0.339; and zeta potential value of -24.2 mV. Enzyme crude extract and purified enzyme of black soybean tempeh at a concentration of 100% gave 51.28 and 56.05% thrombolytic activity. Based on the test results obtained, the optimum formula of photosensitizer liposomes produced is at a good pH, high encapsulation efficiency, with photothermal efficiency of 57.66 and 44.23%, and provides good thrombolytic activity in testing with a 450 nm laser.
