Abstract
Kissing gourami is a local freshwater commodity that is found on the islands of Kalimantan, Java and Sumatra and has the potential to become a superior commodity. Different water characteristics allow kissing gourami to experience changes in genetic characteristics. The purpose of this study was to determine the morphological, morphometric and meristic characteristics, the identity percentage of cyt b gene, genetic distance and phylogenetic and water quality of kissing gourami from Kelekar River and Lebung Ulak Kemang. This research was conducted in November 2024-January 2025. Samples were obtained from Kelekar River and Lebung Ulak Kemang with a total of 10 samples from both locations. The DNA barcoding stage consisted of DNA extraction, DNA amplification using Polymerase Chain Reaction (PCR), electrophoresis, and sequencing of the cyt b gene. The cyt b gene fragment was amplified at 49.9℃ for 30 seconds. The nucleotide length of the cyt b gene in kissing gourami samples 776-778 base pairs (bp). BLASTn of samples HSK 1-5 and HLK 1-5 had similarity of 99.87-100% with Helostoma temminckii from China and Indonesia, 99.36-99.49% from Japan and 99.24-99.48% from Malaysia. All HSK and HLK samples had a genetic distance of 0.000 (0%) with Helostoma temminckii from China and Indonesia and 0.005 (0,5%) with Helostoma temminckii from Malaysia and Japan. Phylogenetic tree construction formed two subclusters with a bootstrap value of 93%. The results of measuring water quality parameters include temperature ranging from 31.3-34.2℃, pH 3.2-6.4, DO 4.4-5.7 mg L-1, TDS 0.14-2.15 mg L-1, ammonia 0.037-0.348 mg L-1, alkalinity 10-70 mg L-1, transparency 12-95 cm.
