Abstract

Penelitian ini bertujuan untuk menganalisis komposisi kimia, aktivitas enzim protease, dan efektivitas antibakteri larutan ekoenzim. Ekoenzim diperoleh melalui proses fermentasi kulit pepaya, belimbing manis, bengkoang, dan jahe selama tiga bulan pada suhu kamar. Setelah fermentasi, ekoenzim disaring menggunakan membran filter 0,22 µm, dilanjutkan dengan pengukuran pH, identifikasi kadar asam, skrining fitokimia, uji kualitatif enzim protease, dan penilaian aktivitas antibakteri menggunakan metode difusi cakram dan penentuan optical density (OD). Hasil penelitian menunjukkan bahwa pH ekoenzim setelah proses fermentasi mencapai 2,27 dibandingkan dengan pH 6,3, yield 70%, dan mengandung total asam 3,11%. Produk ekoenzim terkonfirmasi memiliki aktivitas protease dengan diameter zona bening 28,33 ± 2,08. Ekoenzim mengandung metabolit sekunder, seperti alkaloid, flavonoid, dan saponin. Hasil pengujian antibakteri menunjukkan bahwa ekoenzim dengan konsentrasi 100% (v/v) memiliki daya hambat yang kuat terhadap Staphylococcus aureus, sedangkan konsentrasi 75% dan 50% (v/v) menunjukkan daya hambat sedang, dan konsentrasi 25% (v/v) menunjukkan daya hambat yang lemah dengan metode difusi cakram. Ekoenzim mampu menghambat dan membunuh pertumbuhan bakteri Staphylococus aureus secara berurutan pada konsentrasi 65% (v/v) dan 80% (v/v) dengan metode penentuan optical density (OD)
