Abstract
Parkia speciosa Hassk. leaves contains flavonoids, terpenoids and tannins that have a potency to treat peptic ulcer disease. Parkia speciosa Hassk. Leaves was extracted by maceration method using ethanol 96%. This study aims to determine the effect of polymers in floating mucoadhesive tablet formula with Parkia speciosa Hassk. leaves ethanol extract as the active ingredient. The polymers used are HPMC-K4M as floating polymer and sodium alginate as a mucoadhesive polymer. The combination of the polymer was made in ratio variations HPMC-K4M and sodium alginate F1 (18%: 7.5%), F2 (28%: 7.5%), F3 (18%: 17.5%) and F4 (28% : 17.5%). The analysis includes standardization of extracts, evaluation of granules and tablets quality, drug release profiles shown by DE360, and chemical interaction analysis using FTIR. The test results were then analyzed using Design expert® 8.0.7.1 program. The analysis showed that the HPMC-K4M at high concentrations increases weight and size uniformity, hardness, floating time, and swelling index. Sodium alginate in high concentrations was able to increase weight and size uniformity, floating time,swelling index, and mucoadhesive time. The highest percentage of DE360 is F1 with 29.865% drug release. All tablet followed zero-order with diffusion and erosion mechanism of the polymer matrix. FTIR analysis showed that there is no chemical interaction between the extract and the two polymers used in formulation of floating mucoadhesive tablet. The optimum formula obtained as the ratio of the combination of 18.10% HPMC-K4M and 9.58% sodium.
