Abstract
Cellulose is a complex polysaccharide that is difficult to decompose naturally, but can be degraded by cellulolytic microorganisms through cellulase enzymes. This study aims to isolate and identify cellulolytic bacteria from the water of Taman Firdaus Lake, Sriwijaya University and test their cellulase enzyme activity qualitatively and quantitatively. A total of 17 bacterial isolates were obtained, with 11 isolates showing cellulolytic activity based on clear zone formation on CMC media. Four isolates with the best activity were tested quantitatively using the DNS method, with isolates DTF6 and DTF15 showing high enzyme activity of 3.15 U/mL and 16.17 U/mL, respectively. Two superior isolates (DTF6 and DTF15) were further characterized and identified as the genus Bacillus. The results of this study indicate the potential of local bacteria in producing cellulase enzymes that can be utilized in organic waste management and environmental biotechnology development.
