Abstract

Karst area is an area that can be utilized for the development of land resources, biodiversity, and landscapes. The rock composition of this area has a uniqueness that can be grown by endemic plants. One of them is Hopea celebica Burck. which has the potential as a construction material. Karst areas have fragile ecosystems that are easily impacted by various environmental factors, resulting in many global challenges. This research aims to determine the potential of Hopea celebica Burck. rhizosphere bacterial isolates, the interaction between Hopea celebica Burck. rhizosphere isolates in the process of soil formation and plant growth and identify bacterial species that have the potential to be used as biofertilizer agents in the Maros-South Sulawesi karst areas. The research took place at the Genomics Building Preparation Laboratory 2.4, National Research and Innovation Agency Cibinong-Bogor. The methods used in this study were isolation and purification, biofertilizer potential activity test, compatibility test, and 16s rRNA gene identification analysis. The research identified three bacterial isolates, S4.1, S4.2, and S5.10, with potential as biofitilizer agent from a total of 23 isolates. The species of S4.1, S4.2, and S5.10 demonstrate interaction properties that prevent the formation of inhibition zones between isolates, making them ideal candidates for use as a consortium. The rhizosphere bacteria associated with Hopea celebica Burck. found in karst areas show considerable promise as biofertilizers. They possess the ability to fix nitrogen, solubilize phosphate and potassium, and produce the hormone Indole Acetic Acid based on 16s rRNA gene identification analysis, namely Staphylococcus capitis, Bacillus zanthoxyli, and Serratia marcescens. Kawasan karst merupakan wilayah yang dapat dimanfaatkan untuk pengembangan sumber lahan, keanekaragaman hayati, dan bentang alam. Komposisi bebatuan dari kawasan ini memiliki keunikan yang dapat di tumbuhi oleh tumbuhan endemik. Salah satunya yakni Hopea celebica Burck. yang berpotensi sebagai bahan konstruksi. Namun, dikawasan karst spesies Hopea celebica Burck. telah terjadi penurunan jumlah populasi. Wilayah karst termasuk ekosistem yang rapuh dan mudah terpengaruh oleh beragam faktor lingkungan, sehingga menimbulkan banyak masalah yang dijumpai secara global. Penelitian ini bertujuan untuk mengetahui potensi isolat bakteri rizosfer Hopea celebica Burck., interaksi antar isolat rizosfer Hopea celebica Burck. dalam proses pembentukan tanah dan pertumbuhan tumbuhan dan mengidentifikasi spesies bakteri yang memiliki potensi untuk dijadikan agen biofertilizer pada kawasan karst Maros-Sulawesi Selatan. Penelitian bertempat di Gedung Genomik Laboratorium Preparasi 2.4, Badan Riset dan Inovasi Nasional Cibinong-Bogor. Metode uji yang digunakan pada penelitian ini berupa isolasi dan pemurnian, uji aktivitas potensi biofertilizer, uji kompatibilitas, dan analisis identifikasi gen 16s rRNA. Hasil penelitian yang dilakukan, dari 23 isolat bakteri terdapat 3 isolat dengan kode S4.1, S4.2, dan S5.10 yang memiliki potensi dengan nilai tinggi. Adapun isolat bakteri dengan kode S4.1, S4.2, dan S5.10 memiliki sifat interaksi dengan tidak membentuk zona hambat antrar isolat sehingga cocok dijadikan konsorsium. Spesies bakteri rizosfer Hopea celebica Burck. dari kawasan karst berpotensi sebagai agen pupuk hayati (biofertilizer) yang berfungsi menambat nitrogen, melarutkan fosfat dan kalium, serta menghasilkan hormon Indole Acetic Acid berdasarkan analisis identifikasi gen 16s rRNA yakni Staphylococcus capitis, Bacillus zanthoxyli, dan Serratia marcescens.
