Abstract

Coffea canephora leaves contains alkaloid, flavonoid, and phenolic which are potential for antioxidant activity and cytotoxic. The purpose of this research is to determine activity, cytotoxic, and assay of phenolic total from active extract of robusta coffee leaves. Determination of antioxidant activity was conducted using n-hexane, ethyl acetate, and ethanolic extract with DPPH (1,1-diphenyl-2-picrylhidrazil) method and quercetin as comparison. The result of antioxidant activity study showed that each extract with quercetin comparison have n-hexane value IC50 539.40 g/ml, ethyl acetate 79.64 g/ml, ethanol extract 22.91 g/ml, and quercetin 4.57g/ml. Ethanolic extract as antioxidant activity and the most reactive compare other extracts and stated as active extract. Determination of cytotoxic activity with BSLT method was performed on ethanol extract and has LC50 value 108.14 g/ml. The content of total phenolic used Folin-Ciocalteu reagent method showed total phenolic contents result of ethanol extract was 115.76 mg/g.
