Abstract

Latar Belakang: Adenokarsinoma kolorektal merupakan kanker dengan insidensi kedua tertinggi dan angka kematian ketiga di dunia. Kompleksitas karsinogenesis adenokarsinoma kolorektal dipengaruhi oleh berbagai mutasi gen supresor tumor, salah satunya gen FAT4 yang berfungsi menghambat EMT dan proliferasi sel kanker. Namun, hanya sedikit penelitian yang menyelidiki peran FAT4 pada organ kolorektal, sehingga penelitian ini bertujuan untuk menilai hubungan ekspresi FAT4 dengan grading histopatologi dan stadium klinis. Metode: Penelitian ini merupakan penelitian deskriptif analitik dengan desain cross sectional dan menggunakan 42 sampel blok parafin adenokarsinoma kolorektal. Data klinis berupa usia, jenis kelamin, lokasi tumor, stadium klinis dan grading histopatologi diperoleh melalui rekam medis, kemudian dianalisis menggunakan Chi-square atau fisher’s exact test dan multivariat regresi linear multiple dengan metode backward. Hasil: Terdapat hubungan signifikan antara ekspresi FAT4 dengan grading histopatologi (p 0.014), dimana ekspresi FAT4 yang rendah memiliki risiko grading histopatologi yang tinggi (high grade) sebesar 4.3 kali. Ekspresi FAT4 tidak berhubungan signifikan dengan stadium klinis (p 0.36). Analisis multivariat menunjukkan bahwa usia, jenis kelamin, dan lokasi tumor tidak memiliki pengaruh terhadap ekspresi FAT4 dengan grading histopatologi. Kesimpulan: Ekspresi FAT4 yang tinggi cenderung dijumpai pada adenokarsinoma kolorektal low grade dan sebaliknya.
