Abstract

Dillenia suffruticosa (L.) leaves, commonly known as "Simpur Air", possess therapeutic potential as an anti-hyperuricemic agent. The plant contains secondary metabolites such as alkaloids, terpenoids, flavonoids, and fatty acid esters, which have been shown to inhibit xanthine oxidase (XO), an enzyme associated with hyperuricemia. This study aimed to evaluate the XO inhibitory effect of the ethanolic extract of D. suffruticosa leaves, its anti-hyperuricemic activity, and its effectiveness in reducing interleukin-1β (IL-1β) levels.An experimental study was conducted using a randomized pre-post test control group design with 25 male Wistar rats (Rattus norvegicus) induced with hyperuricemia. The animals were divided into five groups: negative control (Na-CMC), positive control (Allopurinol), and three treatment groups receiving ethanolic extract of D.suffruticosa leaves at doses of 100 mg/kg BW, 200 mg/kg BW, and 400 mg/kg BW, respectively. XO inhibition was evaluated using UV-Vis spectrophotometry, while serum uric acid and IL-1β levels were measured to assess anti hyperuricemic and anti-inflammatory effects. Furthermore, GC-MS analysis was conducted to identify the bioactive compounds responsible for the observed pharmacological activity.The results demonstrated that the ethanolic extract of D. suffruticosa leaves exhibited strong XO inhibitory activity, with an IC₅₀ value of 30.65 ppm. Administration of the extract at a dose of 400 mg/kg BW for seven consecutive days significantly reduced serum uric acid and IL-1β levels (p < 0.05). GC-MS analysis revealed two predominant compounds: hexadecanoic acid (16.09%) and γ-sitosterol (12.82%), both known for their anti-inflammatory and anti-hyperuricemic properties. These findings provide a scientific basis for the development of D. suffruticosa leaf extract as a promising alternative therapy for managing hyperuricemia and associated inflammatory conditions.
