Abstract

Background: Scaffold is a material serves as a place to attach cells and growth factors to mimic the structure of extracellular matrix (ECM), so that it can help restore the function of damaged tissues. Based on their raw materials, scaffolds are divided into two: natural and synthetic. Research continues to be conducted to make plants as scaffolds alternatives as their structure resembles body tissue and retains its shape after the decellularization process. One of these plants is gambier leaves. Gambier leaves have a pore-like structure, similar to porous scaffolds and trabecular bone. The microporosity in scaffolds plays an important role in aiding cell attachment, proliferation and differentiation. Objective: To determine the microporosity of post-decellularized gambier leaves as a scaffold using detergent method. Method: Gambier leaves were cleaned, stored at -20°C, then cut using a biopsy punch. The samples were immersed in 10% Sodium Dodecyl Sulfate (SDS) solution for 5 days, washed with distilled water, and immersed in Tween 20 and NaClO solutions that were changed every 24 hours until the leaves became transparent. The leaves were washed again, immersed in fixation solution overnight, and then dehydrated gradually with ethanol of several different consentration. Samples were dripped with hexamethyldisilazane (HMDS), coated with 50 nm gold, and analyzed by SEM with three magnifications. Result: There were visible wrinkles in the leaf structure due to improper preparation. Conclusion: One of the samples showed the presence of microporosity measuring 0.689 μm and 0.5512 μm. The other sample showed a wrinkled surface due to errors in sample preparation.
