Abstract

Chinese betel leaf (peperomia pellucida) contains secondary metabolite compounds such as flavonoids and terpenoids that have potential as antioxidants. This study aims to determine the effect of giving extracts and fractions of Chinese betel herb invivo on male Wistar rats induced oxidative stress by carbon tetrachloride (CCl4). The 30 test animals were divided into 6 treatment groups, namely the normal group (Na-CMC 1%), negative group (Na-CMC 1% and CCl4), n-hexane fraction treatment group, ethyl acetate fraction treatment group, ethanol fraction treatment group, and total ethanol extract treatment group. Antioxidant activity test was conducted by measuring plasma malondialdehyde (MDA) levels, catalase enzyme activity of liver tissue, as well as macroscopic observations and liver hispatology. The average MDA levels in the normal, negative, n-hexane fraction, ethyl acetate fraction, ethanol fraction, and total ethanol extract groups were 0.969; 2.635; 0.593; 1.208; 1.389 nmol/mL, respectively. The average catalase enzyme activity in the normal, negative, n-hexane fraction, ethyl acetate fraction, ethanol fraction, and total ethanol extract groups were 26.477; 9.385; 37.706; 46.834; 22.231; 15.776 U/mL respectively. The results showed that the administration of ethyl acetate fraction was able to reduce MDA levels significantly (p<0.05) and increase catalase enzyme activity optimally compared to the negative group. Based on these results, it indicates that semi-polar compounds in the ethyl acetate fraction play a role in counteracting free radicals and inhibiting lipid peroxidation.
