Abstract
Refluks laring faring merupakan refluks isi gaster dan duodenum ke regio laring dan faring serta menyebabkan perubahan terkait inflamasi pada regio tersebut. Sejauh ini belum terdapat model hewan yang diterima secara universal untuk memodelkan refluks laring faring. Penelitian ini bertujuan untuk menciptakan suatu model hewan tikus untuk penelitian terkait refluks laring faring sekaligus membuktikan peran refluks laring faring terhadap perbedaan ekspresi IL-6, MUC5AC, dan -catenin pada regio tersebut. Penelitian ini memaparkan pepsin asam sebanyak 0,1-0,2 mL tiga kali sehari selama 5-15 hari secara langsung pada regio laring dan faring. Selanjutnya hewan menjalani sacrifice dan menjalani pemeriksaan ELISA untuk mengukur ekspresi IL-6 jaringan. Ekspresi MUC5AC dan -catenin jaringan dilakukan dengan imunohistokimia. Penelitian ini menemukan bahwa model hewan yang dikembangkan dalam penelitian ini feasible untuk dilakukan. Ekspresi IL-6 laring bersifat time- dan dose-dependent sementara ekspresi IL-6 hipofaring bersifat dose-dependent. Ekspresi MUC5AC laring bersifat dose-dependent. Ekspresi -catenin hipofaring bersifat dose-dependent. Penelitian ini menemukan bahwa terdapat hubungan positif antara ekspresi IL-6 dan ekspresi -catenin dan hubungan positif antara ekspresi MUC5AC dengan ekspresi IL-6. Penelitian ini juga menemukan bahwa terdapat kompensasi pada jaringan laring dan faring terhadap pajanan pepsin asam dengan dosis dan waktu yang diberikan dalam perlakuan ini. Kata kunci: refluks laring faring, model tikus, IL-6, -catenin, MUC5AC Laryngopharyngeal reflux is a condition of gastroduodenal refluxate exposure to laryngopharyngela region, resulting in inflammatory-related changes in these region. Up to this point there is no universally-accepted animal model of laryngopharyngeal reflux. This study aimed to create rat model of laryngopharyngeal reflux in addition to provide evidence of laryngopharyngel reflux-related changes on the expression of IL-6, MUC5AC, and -catenin in mentioned regions. This study directly exposed rat laryngopharyngela region to artificial gastric juice, 0.1-0.2 mL three times per day for 5-15 days. Rat model underwent sacrifice and tissue IL-6 expression was measured through ELISA. Tissue expression of both MUC5AC and -catenin was measured through immunohistochemical study. This study proved the feasibility of the animal model. Expression of laryngeal IL-6 was found to be both time- and dose-dependent while hypopharyngeal IL-6 expression was mainly dose-dependent. Expression of laryngeal MUC5AC was mainly dose-dependent. Expression of hypopharyngeal - catenin was mainly dose-dependent. This study discovered positive correlation between IL-6 and -catenin expressions and positive correlation between MUC5AC and IL-6 expressions. In addition, this study also discovered compensatory mechanism in the laryngopharyngeal region as the result of exposure of the artificial gastric juice in this study.
