Abstract

Cellulase enzymes play a crucial role in various industries, yet challenges related to production costs and purification efficiency remain significant. This study aims to investigate the potential of pretreated rice straw as an alternative substrate for cellulase production from Bacillus licheniformis TS-10, and to evaluate the effectiveness of ammonium sulfate fractionation and gel filtration chromatography in the purification process. Furthermore, the enzyme was characterized based on temperature and pH variations. Enzyme production was conducted via submerged fermentation, followed by a multi-step purification process and activity assays using the DNS method. The results showed that rice straw yielded higher cellulase activity (0.566 U/mL) compared to carboxymethyl cellulose (CMC) (0.366 U/mL), with specific activities of 7.459 U/mg and 5.227 U/mg, respectively. Ammonium sulfate fractionation (20–40%) resulted in a specific activity of 37.807 U/mg with a purification fold of 5.06. Gel filtration chromatography further increased the specific activity, with the highest value observed in Fraction 7 (42.811 U/mg). Enzyme characterization revealed optimal activity at temperatures of 50–60°C and neutral pH levels (pH 6–7), depending on the fraction. These findings suggest that rice straw is a promising alternative substrate, and that the applied purification methods are effective in enhancing the quality of cellulase produced by Bacillus licheniformis TS-10.
