Abstract

The research was isolated secondary metabolite compounds from the n-hexane fraction of laban plant leaves (Vitex pinnata) and conducted cytotoxic tests on shrimp larvae (Artemia salina) which would potentially be used as anticancer drugs. The isolation process was carried out by maceration extraction in methanol solvent, followed by liquid-liquid fractionation using n-hexane and ethyl acetate. Separation of the n-hexane fraction was carried out by Vacum Liquid Chromatography (VLC) and purity testing using TLC with a 3-eluent system. The structure of the isolated compound was determined based on UV and NMR spectroscopy data, while cytotoxic activity was determined by the Brine Shrimp Lethality Test (BSLT) method. The isolated compound was obtained in the form of yellow crystals as much as 10.2 mg. Based on UV and NMR spectroscopy data (1H-NMR and 13C-NMR for 1-D, HSQC and HMBC for 2-D) showed that the isolated compound is a flavonoid compound of the flavonol group, namely the compound 5-hydroxy-3,7,4'-trimethoxyflavone. Cytotoxic test of the isolated compound against A. salina indicated that the isolated compound is cytotoxic with an LC50 value of 46.97 ± 26.03 mg/L.
