Abstract

Tithonia diversifolia is a shrub belonging to the Asteraceae family and one of 12 species classified under Tithonia genus. T. diversifolia is known in Indonesia as kipait, paitan, kembang bulan, and insulin. Previous studies have reported that T. diversifolia contains abundant sesquiterpene lactones compounds. Sesquiterpene lactones have been extensively reported in the aerial parts of T. diversifolia, without separating between stem and leaf parts. This study aims to isolate secondary metabolites from the ethyl acetate fraction of T. diversifolia leaves, followed by cytotoxicity testing. The isolation process involves extraction, fractionation, separation, and purification steps. Extraction was performed by maceration for 1 x 24 hours with methanol as the solvent, followed by liquid-liquid fractionation using n-hexane and ethyl acetate as solvents. Separation and purification of the compounds were carried out using various chromatographic techniques, including vacuum liquid chromatography, radial chromatography, and Sephadex column chromatography. The structure of the isolated compound was determined using UV spectroscopy and NMR (1H-NMR, 13C-NMR, HSQC, and HMBC). The isolated compound yielded 75.1 mg of a yellow solid. Structural elucidation revealed that the isolated compound belongs to the sesquiterpene lactone group with a germacrene skeleton named tagitinin C. Cytotoxicity testing using the BSLT method with Artemia salina larvae at test solution concentrations ranging from 100 to 1.56 mg/L indicated that the isolated compound was cytotoxic with an LC50 value of 37.26 ± 3.03 µg/mL.
