Abstract

This study is conducted to isolate secondary metabolite compounds contain in the ethyl acetate fraction and test the cytotoxic activity of V. pinnata leaf parts. Determination of the structure of the isolate compounds was carried out based on UV and NMR spectrum data (1H NMR, 13C-NMR, HSQC dan HMBC). Cytotoxic activity testing was carried out by BSLT (Brine Shrimp Lethality Test) method against Artemia salina. A total of 5.4 mg of orange-colored solid was isolated from the ethyl acetate fraction of V. pinnata leaves. Based on UV spectrum data, the isolated compound is a phenol derivative. NMR spectrum data (1H-NMR, 13C-NMR, HSQC dan HMBC) showed that the isolated compound is p-hydroxybenzoic acid. Cytotoxic testing using BSLT on methanol extract, n-hexane and ethyl acetate fractions, showed that all three were active cytotoxic with LC50 values of 484.76 ± 25.59; 333.95 ± 35.98; 275.13 ± 36.45 mg/L, respectively. The isolated compound is also cytotoxic active with an LC50 value of 55.33 ± 25.22 mg/L. This data shows that V. pinnata leaf plants and isolated compounds have potential as anticancer compounds.
