Abstract

Asian Redtail Catfish Hemibagrus nemurus is one of species from the genus Hemibagrus which commonly found throughout South-East Asia. The aims of this research is to identify the genetic relation among the Hemibagrus species based on the sequence of gene cytochrome c oxidase sub unit I DNA mitochondria (COI mtDNA) and to construct the phylogenetic tree among catfish species. The research methods used in identifiying species and sequences variation are: DNA isolation, amplification by PCR (Polymerase Chain Reaction) and fragmen COI mtDNA sequencing from the fish samples that were collected from Penukal River at Air Hitam sub-district in Penukal Abab Lematang Ilir (PALI) and Musi river at Arisan Musi sub-district in Muara Enim. The COI gene were sequenced by PCR with optimum annealing temperature 56⁰C for 40s with 30cycles. The result showed Hemibagrus nemurus from Penukal river and Musi river had 572 bp and 596 bp. Based on BLAST (Basic Local Alignment Search Tool) analysis compared to Genbank database, H. nemurus from Penukal river had identity 100% to H. nemurus from Musi river with genetic distance 0,00. It’s claimed to be caused by one of them is the environment that the water quality tends to be same, except in pH parameter in Penukal River which tends to be more acid than Musi river. The habitat characteristics of Asian redtail catfish in Penukal river are temperature (28-29⁰C), pH (7,18-7,58), dissolved oxygen (1,8-3,5 mgL-1), secchi depth transparency (63-98 cm), NH3 (0,11-0,19 mgL-1) hardness (270,5-421,8 mgL-1), while in Musi river are temperature (29-31⁰C), pH (4,4-5,6), dissolved oxygen (3,2-3,5 mgL-1), secchi depth transparency (120-150cm), NH3 (0,09-0,14 mgL-1), and hardness (238,3-392,8 mgL-1).
