Abstract
Halymenia durvillei is a red seaweed species with considerable potential for commercial cultivation in Indonesia. Tissue culture has emerged as a promising method to enhance its biomass production. This study aimed to analyze the most effective combination of plant growth regulators (PGRs) for callus induction and to examine the morphological characteristics of the induced callus. The research was conducted from January to April 2025 using an experimental design supported by descriptive and statistical analyses. Five combinations of PGRs, comprising 2,4-dichlorophenoxyacetic acid (2,4-D), thidiazuron (TDZ), and gibberellic (GA₃), were tested in treatments A through E. All treatments successfully induced callus formation, with effectiveness rates ranging from 73.33% to 100%. Treatment D (2,4-D 0.5 mg/L + TDZ 2 mg/L + GA₃ 0.6 mg/L) resulted in 100% viable callus and achieved the highest average rank based on biological observations, although statistical analysis indicated no significant differences among treatments (Asymp. Sig. = 0.245). The induced callus was white (score 1), compact (non-friable), and microscopic examination suggested it consisted of moniliform filamentous cells.
