Abstract
Cefixime is a broad-spectrum antibiotic with low water solubility, which limits its absorption in the gastrointestinal tract and affects its therapeutic effectiveness. To address this limitation, a submicroparticle formulation of chitosan-alginate loaded with cefixime was developed using the ionic gelation method with CaCl₂ as a cross-linker. This approach aims to enhance the solubility, stability, and encapsulation efficiency of the active compound. The study was conducted with variations in CaCl₂ concentration (20 μL, 40 μL, and 100 μL) to determine the optimal formulation based on encapsulation efficiency percentage (%EE). The results showed that the use of 20 μL CaCl₂ yielded the highest encapsulation efficiency at 90.63%, followed by 83.67% with 40 μL, and 77.17% with 100 μL. Characterization of the best-performing formula revealed a particle diameter of approximately 703 nm, with a PDI of 0.469 and a zeta potential of +31.17 mV, indicating good physical stability of the system. The particles exhibited a spherical morphology and uniform size distribution, suggesting that the formulation process was successful. Physical and chemical stability testing under storage conditions indicated that the formulation remained relatively stable, although further development is needed to ensure long-term stability. The findings of this study are expected to improve the bioavailability of cefixime through the development of an efficient nanoparticle delivery system, thereby enhancing the drug’s therapeutic effectiveness and tolerability.
