Abstract
Antioxidants are compounds that prevent and repair damage to cells in the body, especially those caused by exposure to free radicals. One source of antioxidant compounds is the plant Syzygium myrtifolium Walp. or red shoots. Syzygium myrtifolium is one of the medicinal plant that has been used for the treatment of hypertension, facilitating labour, and abdominal pain. Polyphenolic compounds found in S. myrtifolium are known to act as antioxidants. This study aims to determine the antioxidant activity of fractions from S. myrtifolium leaves, isolate secondary metabolite compounds in the antioxidant active fraction of S. myrtifolium, and identify the components of phenolic derivative compounds present in the antioxidant active fraction. Extraction of S. myrtifolium leaves was carried out by multistage maceration in n-hexane, ethyl acetate, and methanol solvents. Each fraction was tested for antioxidant activity using DPPH. The chemical components in the active fraction were further separated by gravity column chromatography into several subfractions. Chemical profiles were obtained from the identification of pure compounds from the active subfractions by 1H-NMR and 13C-NMR spectroscopic method, and identification of polyphenolic compounds by LCMS/MS. The study found that the ethyl acetate and methanol fractions showed strong antioxidant activity with IC50 values of 99.25 and 93.31 μg/mL, respectively. A pure white solid compound was obtained from the active ethyl asetat subfraction and identified as betulinic acid. Furthermore, the active subfraction of the methanol fraction was analysed by LCMS/MS and identified as containing phenolic derivative compounds such as bergenin, quercetin, elemicin, and acarone.
