Abstract
Free radicals, in the form of reactive oxygen and nitrogen, are formed in the human body through certain physiological processes. Excessive production of free radicals can damage DNA, proteins, membrane lipids, and enzymes, triggering oxidative stress and cell death. Antioxidants play an important role in neutralizing these free radicals. In addition, the increasing resistance of pathogenic microbes to antibiotics, such as Salmonella typhi, Escherichia coli, Bacillus subtilis, and Staphylococcus aureus, has led to the need to search for new antibacterial agents. The Avicennia marina mangrove plant is known to have pharmacological activities, including antioxidant and antibacterial activities. This potential is mostly produced by bioactive compounds produced by endophytic fungi that live on the plant. Endophytic fungi are able to produce secondary metabolites that are similar to their hosts, providing great opportunities for pharmacological exploration without damaging the natural ecosystem. The purpose of the study was to determine the results of isolation and identification of endophytic fungi and to determine the antioxidant and antibacterial activities of endophytic fungal extracts of the Avicenia marina mangrove plant. This research was carried out from March 2024 until completion at the Integrated Laboratory, UIN Raden Fatah Palembang. Plant determination was carried out at the Indonesian Biology Generation Foundation. The research stages include sampling, sample identification, sample sterilization, isolation and purification of endophytic fungi, characterization and identification of endophytic fungi, cultivation and extraction, Antioxidant activity test using the DPPH method and antibacterial activity test using the disc diffusion method and thin layer chromatography test of fungi that have the highest activity. The research resulted in a total of 29 isolates, consisting of 8 isolates from roots, 9 isolates from stems, 6 isolates from leaves, and 6 isolates from fruits. The very strong antioxidant activity was found in isolates AMD2 Diaporthe sp. (19.102 μg/mL), AMF3 Neopestalotiopsis sp. (19.042 μg/mL), and AMF6 Aspergillus sp. (19.678 μg/mL). The antibacterial activity of mushroom extracts isolated from parts of the Avicennia marina plant showed the highest percentage values for isolate AMA4 Diaporthe sp. (84.3%, 83.4%, 85.8%, and 86.6%), isolate AMB3 Aspergillus sp. (81.3%, 80.4%, 85.7%, and 84.7%), isolate AMD2 Diaporthe sp. (84.3%, 83.7%, 85.7%, and 80.2%), and isolate AMF6 Aspergillus sp. (85.3%, 84.4%, 86.7%, and 87.1%) against bacteria E. coli, S. aureus, S. thypi, B. subtilis in succession.
