Abstract

Serbuk gergaji kayu karet mengandung selulosa, yang berfungsi sebagai bahan baku untuk produksi bioetanol. Selulosa dari kayu karet yang telah diolah terlebih dahulu ditransformasikan menjadi gula pereduksi melalui hidrolisis enzimatik menggunakan enzim yang berasal dari Aspergillus niger. Penelitian ini bertujuan untuk mengklarifikasi kinetika Hidrolisis enzimatik selama durasi hidrolisis 5, 10, 15, 20, dan 25 jam pada rasio enzim terhadap substrat 1:20, 2:20, 3:20, 4:20, 5:20, dan 6:20 (v/b). Hasil penelitian menunjukkan bahwa, pada rasio enzim terhadap substrat 6:20 (v/b) dan waktu hidrolisis 25 jam, jumlah maksimum gula pereduksi adalah sekitar 3,15 g L−1. Aktivitas FPase adalah 255,1672 U mL-1, sedangkan aktivitas CMCase mencapai 235,9105. Model kinetik Michaelis–Menten menyatakan bahwa laju maksimum berkisar antara 6,50×10−7 hingga 2,6×10−6 Mol L−1 s−1, dan laju reaksi menghasilkan konstanta Michaelis antara 0,0051 dan 0,0055.
