Abstract

Rifampicin is an antibiotic bactericide against Mycobacterium tuberculosis which has capability to grow in lungs macrophages during oxidative condition. The submicro particles of rifampicin has been produced to increase the stability of rifampicin from oxidative condition in lungs macrophages. Preparation of rifampicin in the form of submicro particles consisted of PLGA as polimer which are stabilized by PVA using emulsion solvent evaporation method. Formulation submicro particles of rifampicin, PLGA, PVA, and ethyl acetate using ultra turrax® speed variation at 13000 rpm, 14000 rpm, and 15000 produce an encapsulation efficiency percentage (%EE) 86.17 ± 0.65, 78.73 ± 1.41, and 66.8 ± 1.4 respectively. The first formula with speed 13000 rpm produced the highest EE percentage which chosen as the optimum formula. The higher value of EE percentage showed more rifampicin has been encaptulated. The result of submicro particles characterization such as diameter and particles distribution (Poly dispersity index/PDI), and zeta potential using particle size analyzer (PSA) on optimum formula 339.6 nm, 0.341, and -38.8 mV. Morphology particle of optimum formula was observed by using transmission electron microscopy (TEM) showed spheric particle and PDI value 0,72. The result of stability studies from optimum formula (F1) such as observations of turbidity and EE percentage produce less significant observations and the value of EE percentage has been changed from 87.06 % ± 1.02 to be pH 5.6 were 46.16% ± 1.35, pH 6 were 66.4% ± 1.01 and pH 7 were 74.9% ± 1.6. Based of statistical analysis independent t-test generate sig. value larger than 0.05 that means the speed of ultra turrax in F1 and F2 does not affect particle size distribution of the formulas.
