Abstract

Swamp buffalo (Bubalus bubalis Linn.) Pampangan a buffalo varieties of native and one of the genetic wealth in South Sumatra with a distribution area in the district Pampangan (Ogan Ilir and Ogan Ogan Ilir) and Banyuasin. The purpose of this study to determine genetic variation and kinship 4 variants Pampangan swamp buffalo. The method used by molecular approaches PCR-RAPD using primer 5 is ILO 1204, ILO 1212, ILO 525, OPW OPY 03 and 13. Data were analyzed using NTSYS program Ver 2.1. Based on the results of amplification, all primary produce 100% ribbon with a total of 20 polymorphic DNA bands with an average every primary produces 4 DNA. Primer that generate the most polymorphic DNA bands are OPW 03 (6 DNA). Based on these results,Genetic variation Pampangan swamp buffalo (Bubalus bubalis Linn.) Striped variant, Red, and Lampung included in the category of close unless genetic variation marked the swamp buffalo Black more band formed on the amplification of polymorphic DNA, Kinship between variants Pampangan swamp buffalo (Bubalus bubalis Linn.) Is low with 0.46 similarity coefficient values (less than 1). Dendogram kinship between variants Pampangan swamp buffalo (Bubalus bubalis Linn.) Dividing the 2 groups (clusters) variants, cluster 1 consists of a buffalo Black, and cluster 2 consists of buffalo Lampung, Red and Stripe.
