Abstract

There are three common commercial tilapia species Oreochomis niloticus, O. mossambicus, and O. aureus. These species have the opportunity for hybridization. Hybridization between tilapia species is widespread, one of which is red tilapia, is a crossbreed of four different species of the genus Oreochromis, namely O. mossambicus, O. niloticus, O. hornorum, and O. aureus. The purposes of this study were determine the morphological, morphometric, and meristic characteristics of farm Nile tilapia and wild Nile tilapia in Toba Lake, to analyze of species or strain composition of nile tilapia using KASP assay PCR, and to determine the physical and chemical characteristics of Nile tilapia habitat. This research was conducted in November 2024- February 2025. Fish and water samples were taken from Toba Lake. The SNP marker stages included DNA extraction, electrophoresis, and SNP marker diagnostics. The analysis results showed the species composition of farm and wild samples, indicated a difference in diagnostic homozygotes. Individuals from the farm population were consisted mainly of O. niloticus species with a diagnostic allele frequency of 0.64, with contributions from non-O. niloticus with an alternative allele frequency of 0.36. Meanwhile the wild population in Toba Lake showed that five individuals were heterozygous with a diagnostic allele frequency value of 0.5 and an alternative allele frequency of 0.5. The water quality parameters of the two locations included temperature of 25-26°C. pH 7.5-8.5. Dissoleved Oxygen 5.01-9.50 mg L-1. TDS 0.45-0.57 mg L-1. alkalinity 50-60 mg L-1. ammonia 0.45-0.57 mg L-1. transparency 0.89-1.9 m.
