Abstract
Thrombosis is a condition in which a blood clot (thrombus) forms in a blood vessel. Treatment involves antithrombotic agents such as anticoagulants, and researchers developed silver-magnetite nanoparticles (Ag-Fe₃O₄) using a green synthesis method using robusta coffee bean extract (Coffea canephora) containing chlorogenic acid as a natural reducing, stabilizing, and thrombolytic agent. The aim was to evaluate the effect of varying concentrations of coffee extract and PEG (6000) acting as a coating agent on temperature, photothermal efficiency, thrombolytic activity, and stability in phosphate buffer saline (PBS) and blood. Optimization was carried out using a 22-factorial design with extract concentrations of 0.5% and 1%, while PEG was 10% and 20%. The optimum formula was obtained at 1% PEG 10% extract, which produced amorphous nanoparticles with a non uniform distribution and a particle size of approximately 50 nm. Characterization using TEM showed results in the form of irregular amorphous and FTIR successfully identified OH, C-H, C=O, C-C and -COOH groups which aimed to demonstrate the success of synthesis and encapsulation in nanoparticles. Performance tests showed that the nanoparticles were able to increase the temperature, where the addition of PBS helped maintain temperature stability but reduced heating efficiency. Meanwhile, without PBS the temperature achieved was higher but tended to be unstable. Photothermal efficiency was recorded at 39.21% accompanied by a high level of hemolysis, but still showed significant thrombolytic activity when tested using a laser at wavelengths of 450 and 550 nm.
