Abstract
Ergocalciferol is used as a medicine to treat rachitis that classified in the Biopharmaceutics Classification System (BCS) class IV. Low solubility and permeability of ergocalciferol will reduce drug bioavailability and affect the absorption. This research aims to increase solubility and permeability of ergocalciferol, so the drug can be applied topically. Ergocalciferol was formulated by single emulsion solvent evaporation method into 3 formulas (1.6; 2.0; and 2.4%) using Poly(Lactic-co-Glycolic Acid) (PLGA) as polymer and Poly(Vinyl Alcohol) (PVA) as stabilizer. Determination of entrapment efficiency percentage (%EE) for formula 1 was 75.92 ± 0.735%, formula 2 was 67.96 ± 0.790 and formula 3 was 53.8 ± 2.097%. Based on result of percent EE, the optimum formula of submicron particle PLGA-ergocalciferol is formula 1 that contain 40 mg PVA. The result of submicro particles characterization such as diameter and particles distribution (Poly Dispersity Index/PDI) and potential zeta using Particle Size Analyzer (PSA) were 270.7 nm; 0.208; and -0.2 mV. Morphology particle of optimum formula was observed using Transmission Electron Microscope (TEM) showed spheric particles which were homogen with a PDI of 0.15. There was a difference in the in-vitro diffusion test results by using Franz diffusion cell, PLGA-ergocalciferol submicro particles was 23.305 ± 0.118%, compared with pure ergocalciferol solution with diffusiom percentage was of 9.381 ± 0.008%.
