Abstract

According to Biopharmaceutics Classification System (BCS), roxithromycin is classified in class II which means it has low solubility in water but high permeability towards biological membrane. Low solubility will affect drug‟s bioavailability hence, preparation and characterization of submicro roxithromycin has been done to improve the solubility in order improve the bioavailability in systemic circulation. Preparation has been done with single emulsion solvent evaporation method using Poly (Lactic-co-Glycolic Acid) (PLGA) as polymer and Poly(Vinyl Alcohol) (PVA) as stabilizer. PVA‟s concentration was varied in 3 formulas (1,6; 2,0; and 2,4 %). Determination of encapsulation efficiency percentage (% EE) for formula 2 was 95,697 ± 0,093 % whereas the value for formula 1 and 3 were 94,831 ± 0,165 and 95,507 ± 0,137 %.Characterization of diameter, polydispersity index (PDI), and zeta potential using Particle Size Analyzer (PSA) for formula 2 were 288,2 ± 27,4 nm, 0,081, and -1,3 mV, while the result of formula 3 were 253,3 ± 11,8 nm, 0,047, and -0,5 mV.Morphology particle analysis of formula 2 has been done using Tranmission Electron Microscope (TEM) which showed spheric and rod shaped particle. Stability test of its physical properties using variation of pH for 14 days showed formula 2 was more stable in pH 6, but not in pH 7. Dissolution test which has been done for submicro PLGA-roxithromycin and roxithromycin conventional capsule obtained the result of Dissolution Efficiency (DE) 82,939 ± 2,298 % and 3,466 ± 0,101 %. Respectively rate and mechanism of release PLGAroxithromycin followed zero order and Higuchi model. The DE value showed that submicro dosage form could improve roxithromycin solubility which resulted towards higher bioavailability compared to conventional dosage form.
