Abstract

The jumputan cloth industry is a home industry that generally used dye as the primary material in the manufacturing process. Dyes are generally used is a synthetic dye. Utilization of synthetic dyes in the dyeing process results in dye liquid waste which will have a negative impact because it is difficult to degrade in the environment. Therefore, a proper treatment system is needed for degrade the textile dye. One of the biological agents that can be used is fungi. According to some previous studies indigenous fungi can be used as agent for dye degradation of textile dye. Therefore, it is necessary to isolate and identification the fungi that can degrade the textile dyes from the industrial waste of jumputan craftsman. The aimed of this research was to find fungi isolates and determined taxon of fungi isolate from industrial waste of jumputan craftsman that can degrade four dye tested. This Study was conducted in December 2016 until June 2017 at Genetics and Biotechnology Laboratory, Microbiology Laboratory, Biology Department Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The sampling process was done by stratified random sampling method. Screening of textile-dye degrading fungi was done by using the agar plug method on solid media. Characterization of fungi was done macroscopically using PDA, MEA, CDA and microscopically using preparation and Henrici's slide culture methods. The results showed that from 21 isolates obtained, 4 isolates i.e. isolates A2P5J2,A3P6J1, TRP5J1, TRP6J2 can degrade four dye tested i.e. direct blue, direct bordeaux, direct green and direct yellow. Positive isolates can degrade dye characterized by the presence of decolorization zone and discoloration around the media. Isolates A2P5J2, A3P6J1, TRP5J1, TRP6J2 have different macroscopic and microscopic characters, therefore these four isolates are suspected to be four distinct species. Conculusion of this research are 4 isolates of fungi can degrade direct blue, bordeaux, green and yellow dyes with the isolate code A2P5J2, A3P6J1, TRP5J1, and TRP6J2. Characterization result show that 4 isolates of fungi approaching characteristic Ascomycetes and Basidiomycetes class.
