Abstract

Uncaria gambir Roxb. ethyl acetate leaves extract contains polyphenol catechin as antibacterial, antiinflamation, and antioxidant used for gingivitis remedy. Formulation of gingival mucoadhesive patch aims to improve the effectivity and prolong residence time. Mucoadhesive patch was formulated by chitosan and PVA as factors used 22 factorial design Design Expert® 10 program. The influence of factor composition and its interaction observed by the analysis result of weight and thickness uniformity, surface pH, folding endurance, percent elongation, swelling index, and mucoadhesive time. Result were analyzed by DX® 10 to determine the optimum formula. Optimum concentration of 3% chitosan and 1% PVA had RSD weight uniformity values of 1.482 ± 0.328% and thickness of 3.012 ± 0.516%, surface pH values of 5, folding endurance of >300 folds, percent elongation of 49.999 ± 3.333%, swelling index of 54.870 ± 9.777%, and mucoadhesion time of 365.88 ± 13.700 minutes. Chitosan decreased weight and thickness uniformity, but increased percent elongation, swelling index, and mucoadhesive time. PVA increased mucoadhesive time, but decreased weight and thickness uniformity, percent elongation, and swelling index. Phenolic content of optimum formula of 18.54 ± 0.023%. Measurement of %diffused optimum formula and pure solution of extract on the 360th minutes (6 hours) are 16.9356 ± 0.218% and 22.6472 ± 0.277%. Compartmental analysis was observed using WinSAAM® resulting optimum formula follows lag compartment model with correlation between Qo and Qc proven by p-value of <0.05. Study of phenolic content stability using heating/cooling method at 4 and 40oC every 24 hours during 7 days show that optimum formula more stable compared pure extract. FTIR test results show that there is no chemical interaction between Uncaria gambir Roxb. ethyl acetate leaves extract, polymer, and plasticizer.
