Abstract

Preparation of dexamethasone into ethosome scale aims to resolve the permeability problem (biopharmaceutical classification system III) and to increase penetration through the skin. Cooling method is used in preparing dexamethasone encapsulated by soy lechitin, propylene glycol, and ethanol 96%. The optimization of dexamethasone ethosome formula with factorial design 23 in Design Expert®10 software is useful to determine the effect of soy lechitin, propylene glycol, and ethanol 96% factor on pH, viscosity, and percent efficiency encapsulation (%EE) as responses. The proportion optimum formula components obtained were 1% soy lechitin, 10% prpopylene glycol, and 20% ethanol 96% concentration with the response values which resulted pH of 6.42, viscosity of 1.26 cP, and percent efficiency encapsulation (%EE) of 90.56%. The optimum of soy lechitin, propylene glycol, and ethanol 96% were known to increase percent efficiency encapsulation (%EE). The resulting diameter, PDI (poly dispersity index), and zeta potensial analysis of the optimum formula were 352.3 nm, 0.441, and -20.7 mV. Particle penetration testing has been done through in vitro diffusion test, using the Franz Diffusion Cell. The diffusion test showed the highest diffusion percent value on dexamethasone ethosome (92.95% ± 0.23) compared with pure dexamethasone (47.72% ± 0.32), and dexamethasone cream (82.17% ± 1.83). The compartement analysis using WinSAAM® program showed that optimum formula followed 2 compartement model. Stability of dosage form was tested through heating cooling cycle method showed a decrease of significant level in each cycle. Ethosome produced have good characteristics but have a poor stability.
