Abstract

Chlorophyta is the largest group of algal vegetation that has chlorophyl pigments for photosynthesis. Molecular DNA is used to distinguish the type of chlorophyta originally from swamp water and others water resources. Rapid identification is PCR techniques performed to sequence DNA and their components in phylogenetic trees. Swamp fish cultivation needs to be supported by a typical microalgae for natural feed, and green water system. The purpose of this research is to know the sequences nucleotide at 18 rRNA genes on chlorophyta isolates from swamp waters and catfish culture pond. This research was carried out at the Laboratory of Plant Physiology and Basic Fisheries Laboratory, Aquaculture Study Program, Faculty of Agriculture, Sriwijaya University from February to August 2018. Primer used in DNA identification is Fw_ITS1 and Rv_ITS 4, which resulted 873 bp. Chlorophyta DNA sequence from this study was contructed as phylogenetic tress with sequence from Gene Bank using BLAST program and Mega 6 maximum likelihood method. The results of the BLAST analysis showed that Chlorophyta isolates from ponds and swamps had proximity to Micractinium sp. LBA 32 (Brazil) with a match percentage of 91% and 97%.
