Abstract

Soybean (Glycine max L.) is a plant that can be used for consumption and medicine mainly in Asia. Soybean seeds contain several active and bioactive compounds, namely flavonoids, isoflavones and antioxidants. The dosage formulation was preceded by coating ethanol extract of soybean seeds with chitosan and sodium alginate polymers so that it could change the particle shape of soybean seed extract (Glycine max L.) into a submicro particle preparation in order to increase the penetration of preparations into the skin. Chitosan is a cationic natural polymer. The cationic features make chitosan able to interact with anionic compounds through crosslink and form particle in submicro size. It is known that flavonoid levels in soybean seeds are 0.104±0.0036%. Submicro particle components consist of soybean, seed extract, chitosan, sodium alginate, and CaCl2 using variations of CaCL2 of 2.5; 4.5; and 6.5 ml. The three formula produce percentage encapsulation efficiency (EE) of 90,71%±0,001; 89,86%±0,001%; and 88,05%±0,001% respectively. The results of submicro characterization of particles such as diameter and particle distribution (Poly Dispersity Index/PDI), and potential zeta using a particle size analyser (PSA) at the optimum formula were 1379,137 nm; 0.396; and 4.8 mV. The results of the X-Ray Diffraction (XRD) analysis of the optimum submicro formula of the particles showed a change in the amorphous form characterized by a decrease in the maximum lamda peak. Submicro preparations of soybean seed extracts are tested by heating cooling cycle with a variation of pH 2.0; 5.5; and 7.4. Testing at pH 5.5 and 7.4 is relatively good while at pH 2.0 there is a drastic decrease in flavonoid levels.
