Abstract
Spirulina production technology in catfish waste media can be an alternative to reduce environmental pollution. However, some environmental factors such as nutrition, light and water content can influence the morphological shape of Spirulina and variations at the genetic level. DNA barcoding can be used to identify species and determine DNA characterization and genetic variation between species. The purpose of this study was to identify the species and morphological forms of Spirulina that were cultured using technical fertilizer and waste media, as well as to know the phylogenetic trees between species from the results of the research and the GeneBank center based on the 16S rRNA gene. This research was conducted at the Aquaculture Laboratory, Aquaculture Study Program, Microbiology and Biotechnology Laboratory, Fisheries Product Technology Study Program, Department of Fisheries, Plant Physiology Laboratory, Agronomy Study Program, Faculty of Agriculture, Sriwijaya University at December 2018-February 2019. The method used in this study DNA isolation, amplification using PCR (Polymerase Chain Reaction) and sequencing with 16S rRNA gene fragments. This study described the morphology of Spirulina cultured in technical fertilizer (ST) and waste (SL) media, genetic distance and phylogenetic tree of Spirulina which were constructed using the Neighbor Joining (NJ) method. The results showed that the isolate used in the study was Arthrospira. SL isolat has linear and helical filaments but the size was shorter than ST. The genetic distance of ST and SL isolates was 0.068 (6.8%). Phylogenetic trees showed that Spirulina platensis was cultured in technical fertilizer media (ST) had a close relationship with Arthrospira platensis petH from Japan (bootstrap value of 95%). Spirulina platensis cultured in waste media (SL) formed a separate subcluster of ST isolates and Arthrospira platensis petH from Japan (bootstrap value of 85%).
