Abstract

Diabetes mellitus is a disease in when the body cannot use insulin effectively. Acarbose has been used to treat diabetes mellitus by inhibiting the action of the α-glucosidase enzyme. Base on literature studies no reports of karamunting stem (R. tomentosa) as α-glucosidase inhibitors. Based on this the isolation of the compounds was carried out and the activity of the α-glucosidase enzyme was tested against extracts, fractions, and pure compounds from caramunting stem. The isolation process was carried out by maceration using as the methanol solvent, followed by liquid-liquid extraction with n-hexane, ethyl acetate, and n-butanol. Separation and purification were carried out on the ethyl acetate fraction by vacuum liquid chromatography and gravity column chromatography. The isolated compound was obtained as a yellowish-white solid that decomposed at 182 °C. Based on the analysis of the UV, IR, 1H-NMR, 13C-NMR, HSQC and HMBC spectra as well as the comparison of literature data that the isolated compound was 3β-O-E-4-hydroxycinnamoyloleanolic acid. Citations : 57(1986-2017) Diabetes mellitus merupakan penyakit dimana tubuh tidak dapat mengunakan insulin secara efektif. Akarbosa telah digunakan untuk mengobati diabetes mellitus dengan cara menghambat kerja dari enzim α-glukosidase. Studi terakhir belum ada laporan tentang batang karamunting (R. tomentosa) sebagai inhibitor enzim α-glukosidase. Berdasarkan hal tersebut dilakukan isolasi senyawa dan pengujian aktivitas enzim α-glukosidase terhadap ekstrak, fraksi, dan senyawa murni dari batang karamunting. Proses isolasi dilakukan secara maserasi menggunakan pelarut metanol, lalu dilanjutkan dengan ekstaksi cair-cair dengan pelarut n-heksana, etil asetat, dan n-butanol. Pemisahan dan pemurnian dilakukan pada fraksi etil asetat dengan Kromatografi cair vakum dan kromatografi kolom gravitasi. Senyawa hasil isolasi didapatkan berupa padatan berwarna putih kekuning-kuningan yang terdekomposisi pada suhu 182°C. Berdasarkan analisa dari spektrum UV, IR, 1H-NMR, 13C-NMR, HSQC dan HMBC serta perbandingan data literatur bahwa senyawa hasil isolasi adalah asam 3β-O-(E)-4-hidroksilsinamoiltoleanat. Ekstrak metanol, fraksi etil asetat, n-butanol mempunyai kemampuan penghambatan α-glukosidase lebih kuat dibandingkan dengan akarbosa, sedangkan senyawa hasil isolasi lebih lemah dari akarbosa.
