Abstract

Vernonia amygdalina have been used traditionally for the treatment of diabetes and other diseases related to antibacterial activity. Information on the chemical compounds and biological activity of this plant has been widely reported but especially from non polar extracts are still limited. This research aims to isolate secondary metabolites from the active antibacterial extract the leaves of V. amygdalina. The research was initiated by used maceration method extraction using gradient polarity solvents, then antibacterial activity of each was tested by disk diffusion method with test bacteria Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Shigella dysenteriae. Separation and purification of antibacterial activity extract was carried out using chromatographic techniques. The pure compound obtained were tested antibacterial activity and structural determination by spectroscopic data analysis of the UV-Vis, IR, 1H NMR, 13C NMR, DEPT 135, HMBC, and HSQC spectra as well as comparised of data in the literature. From n-hexane extract, pure isolates were obtained in the form white crystal (15 mg). Pure isolate at a concentration of 1.000 ppm showed moderate antibacterial activity against the S. aureus with a diameter of 7.4 mm. Based on spectroscopic data analysis, pure isolates is stigmasterol. Stigmasterol is a compound that was first reported from the n-hexane extract the leaves of V. amygdalina.
