Abstract

Gambir water extract. contains catechin compounds with activities as anti-inflammatory, antibacterial, and antioxidants that can be used for gingivitis therapy in the preparation of gingival mucoadehesive patch Water extract of fambir leaves was used as an active substance in the preparation of gingival mucoadhesive patches to prolong residence time. The formulation used a combination of polymer carrageenan and tapioca starch as a factor with a factorial design 22 in the Design Expert®10 program. The responses observed to the effect of factor concentration and interactions were weight, weight uniformity, surface pH, folding endurance, % elongation, swelling index, and mucoadhesive time. The test results for each response were carried out by DX®10 analysis to obtain the optimum formula. The concentration of carragenan 2% and 1% tapioca starch produced the optimum formula with a desirability value of 0.884. The value of % CV of uniformity of weight was 0.9495 ± 0,0005%, CV thickness uniformity was 0.9738 ± 0.005%, % elongation was 53.333 ± 5.773%, swelling index was 55.8512 ± 1.0258%, pH surface 6, folding endurance > 300, mucoadhesive time 356.333 ± 5,5075 minutes. Carrageenan decreased CV weight uniformity, thickness, percent elongation but increased percent development and mucoadhesive time. Tapioca starch decreased CV weight and thickness uniformity, percent development but increased percent elongation and mucoadhesive time. The content of catechins in mucoadhesive patch was 91.7589 ± 0.1858%. Diffused % in patch and exctract were 51.1084 ± 0.0556% and 66.6597 ± 0.655%. Compartmental analysis was observed using WinSAAM® resulting in pure extract and optimum formula following the 3 compartment model with the correlation between Qo and Qc with p-value <0.05. Stability testing of the content by heating /cooling methods in 3 cycles showed that the optimum formula was more stable than pure extract with a decreased in percentages of 1.1087 ± 0.1650% and 1.9918 ± 0.0065%. FTIR test results showed no chemical interaction between gambir leaf water extract with polymers and plasticizers.
