Abstract

The purpose of this study was to find out the specific alleles of each oil palm origin, the kinship between populations resulting from crossing Tenera x Pisifera from different origins, polymorphic SSR markers on the Tenera x Pisifera crossing in the Ghana, Niegeria and Dami oil palm origin. The study began from September to December 2018 at the Integrated Laboratory of PT. Sampoerna Agro Tbk., Palembang. The genetic material used is 3 populations (144 oil palm) Ghana, Nigeria and native/dami palm oil planted in Surya Adi, Mesuji. DNA sample was extracted using the protocol kit of Promega Kit Wizard® Genomic DNA Purification Kit. A total of 30 SSR markers were used in primary screening and 16 markers were selected for the genotype analysis. The results of the Polimorphis Information Content (PIC) analyzed using CERVUS had average value 0.647 which were medium informative level. The number of alleles detected from each SSR marker ranged from 3-6 alleles (mean = 4.625). Genetic distance dendogram was constructed by Unitited Pair Group Method Arithmetic (UPGMA) using NTSYS 2.0pc software (Numerical Taxonomy and Multivariate Analysis System) in 144 oil palms which divided into 2 large groups with genetic similarity coefficients of 0.21 or 21%. Phylogenetic analysis using DARwin 5.05 and Principal Coordinating Analysis (PCoA) using GenALEx ver 6.5 indicated that 3 oil palm populations presented in 3 groups based on origin. The results of Molecular Analysis Variance (AMOVA) showed that the diversity between populations of 43% for diversity between individuals was only 2% then the diversity of individuals in the population was 55%.
