Abstract

Hydrocortisone acetate is low potency class VII topical corticosteroid, due to its hydrophobic and low penetrating properties. In this research, hydrocortisone acetate was modified into ethosome to enhance the topical penetrating property to systemic circulation. Ethosome was chosen to enhance solubility of hydrocortisone acetate, so the entrapment efficiency of vesicle can be enhanced. Hydrocortisone acetate ethosome was prepared by cold method because the potency and effectiveness of hydrocortisone acetate can decrease by high temperature. Experimental design that was used in this research is 23 factorial design, which is the two level full factorial design for three factors. The factors in this formulation were soy lecithin, propylene glycol, and ethanol 96%, while the levels were minimum and maximum. The variation of concentration were soy lecithin (in 1 and 4%), propylene glycol (in 5 and 10%), and ethanol 96% (in 20 dan 40%). The optimized formula consisted of 1% soy lecithin, 5% propylene glycol, and 20% ethanol 96%. pH value, viscosity, and %EE of the optimized formula were 6.62; 1.91 cP; dan 85.37% respectively. The result of morphological analysis on optimized formula was spherical particle with PDI value 0.25. Particle size, PDI value, and zeta potential of the optimized formula were 209.6 nm; 0.493; and -13.7 mV respectively. There was no new infrared peak observed in placebo ethosomal vesicles, hydrocortisone acetate loaded ethosome, and hydrocortisone acetate, so there was no chemical interaction between components. The results of assay determination at cycle 6 of hydrocortisone acetate loaded ethosome, 1% hydrocortisone acetate cream, and hydrocortisone acetate were 29.4691%; 22.9053%; and 34.3459% respectively, while the percent of diffusion achieved within the first 12 hours were 53.16887%; 45.4850%; and 32.0767%. Based on the results of characterization data, the resulting ethosome has good characteristics and is feasible to develop, but tends to be less stable due to the lysis of the vesicles.
